The criteria that should be adopted in the selection of cells for examination by LLS will be those which lead to the minimum heterogeneity within the scattering volume. This is most readily achieved in tissues of one or a few cells in thickness, or in single cell systems. Many unicellular systems have a cytoplasmic structure which, while not rigidly stratified, at least allows the selection of areas rich in particular organelles, and areas of activity specifically related to certain functions. Some of these systems will be considered at this meeting.
